In his July 2005 professional debut, Khan scored a TKO victory over David Bailey. He collected three more wins that year, six more in 2006 and stopped Mohammed Medjadji in his first appearance of 2007. Against Stefy Bull that April, Khan made his HBO debut, winning by technical knockout in three rounds. He earned three more victories to finish the year 5-0.

Khan won a unanimous decision and produced two knockout wins to begin 2008. That September, he met undefeated Breidis Prescott, suffering the first loss of his career. Three months later, he bounced back with his 15th knockout win against Oisin Fagan.

In his first appearance of 2009, Khan defeated Marco Antonio Barrera. Moving up to the 140-pound division, he claimed his first world title with a unanimous decision win over Andriy Kotelnik in July. In his first defense, Khan stopped undefeated Dmitriy Salita in the first round of their December contest.

In May 2010, Khan knocked out Paulie Malignaggi in his first fight outside of the United Kingdom. He defeated Marcos Maidana by unanimous decision that December in his fourth consecutive defense. Khan won against Paul McCloskey in April 2011 and knocked out 140-pound world titlist Zab Judah at Mandalay Bay in July. He faced Lamont Peterson five months later, losing a split decision.

With another shot at a 140-pound world title in July 2012, Khan lost to undefeated Danny Garcia. That December, he stopped undefeated Carlos Molina in ten rounds and defeated Julio Diaz in his sole appearance of 2013.
